The Core supplies viral-based RNAi constructs targeting every gene in the human and mouse genomes. The DNA is transfection quality which can be delivered directly to target cells or used to create infectious lentiviral constructs.

Constructs are ordered by gene and in general there are 5 constructs per gene. On average two or more constructs will knockdown their target genes by greater than 75%.

Core Description

The Core supplies viral-based RNAi constructs targeting every gene in the human and mouse genomes.  The DNA is transfection quality which can be delivered directly to target cells or used to create infectious lentiviral constructs.

Constructs are ordered by gene and in general there are 5 constructs per gene.  On average two or more constructs will knockdown their target genes by greater than 75%.

RNAi DNA Request Procedure:

The Gene Symbol and Gene ID of interest should be sent via email to the following address:

rnai-request@gowustl.onmicrosoft.com

Please specify whether you would like human or mouse clones.  Upon receipt of the request, we will send you an official cost estimate for the distribution of the clones. Estimates can be accepted via email with a simple reply from you, by fax, or in person.  Instructions will be sent with the cost estimate.

Once the estimate has been officially accepted, DNA will be provided to you.  For each gene request, all available hairpin constructs that target the gene of interest will be supplied in a 96 well format.  Each sample will consist of ten microliters of sample.  In addition, a template in the form of a table will be supplied with each request.  This template will list the locations of each construct.  Control wells will be provided in the following manner.  Well A1 of the container will be left empty as the negative control.  Wells A2 through A5 will contain a single construct that targets RFP, GFP, LacZ, and Luciferase, respectively.

The quality of each sample will be confirmed by visualization on an agarose gel.  If the research performed with these clones leads to a publication, the requestor is required to acknowledge the TRC, CDI, and the MGI.

Pricing

Pricing is subject to core verification

In general, there are 5 constructs per gene. Each construct costs $30.00, so each gene will cost approximately $150.00.
